Background: Bone marrow is the soft material found inside most large bones of the body. Bone marrow produces red blood cells, white blood cells, and platelets that are released into the blood stream. Inside the marrow, these cells start off as young, immature cells called progenitor cells. Researchers want to study these cells in healthy people. Objective: To understand how progenitor cells change with age. Eligibility: Healthy people ages 18 and older Design: Participants will be screened with a questionnaire, a physical exam, and blood tests. Participants will have a bone marrow aspirate. They will be asked to lie on their stomach or side. A local anesthetic will be injected with a small needle under the skin at the site. A needle will then be placed through the skin and into the hip bone. A small amount of the liquid part of the bone marrow will be taken up into the needle. After the needle is removed, a pressure bandage will be placed on the site. Participants will be asked to become a repeat volunteer and have a bone marrow aspirate sample collected once every other year. They will have a physical exam and blood tests before each collection. The cells from the collection will be used for genetic testing and research. Participants will be in the study for as long as they remain healthy and willing to participate.

Eligibility Criteria

Eligible

You may qualify if you...

  • Healthy males and females age 18 years and older
  • Agree to DNA/RNA extraction, analysis, and storage of collected samples
  • Health history questionnaire shows no active infection or HIV risk
  • Able to read and speak English
  • Willing and able to come every 2 years for bone marrow aspiration procedure at NIA Clinical Research Unit
Not Eligible

You will not qualify if you...

  • Unable to provide valid government-issued identification
  • Unable to provide informed consent
  • Allergy to lidocaine used during bone marrow aspiration
  • Positive test results for viral infections like RPR, HIV, Hepatitis B or C (unless treated Hepatitis C with undetectable RNA)
  • Ongoing risk factors for HIV or Hepatitis B or C, including intravenous drug use or risky sexual contact
  • Major medical illnesses such as cancer, deep vein thrombosis, autoimmune disorders, organ or bone marrow transplant, or serious liver, kidney, heart, or lung disease
  • Medical conditions that make the procedure unsafe (e.g., Parkinson's disease, dementia, uncontrolled behavior)
  • Bleeding disorders like hemophilia or von Willebrand's disease
  • Significant abnormalities in blood tests such as elevated liver enzymes or low kidney function
  • Taking medications that affect blood cell function such as chronic steroids, antivirals, or chemotherapy
  • Past or present radiation therapy
  • Conditions preventing access to the iliac crest (wounds, rashes, large fat deposits)
  • Organ, tissue, bone, or skin transplant
  • Pregnancy or nursing within the last 6 weeks (temporary deferral)
  • Recent use of certain medications or vaccines (with specified deferral periods)
  • Recent infections, fever, or surgical procedures (with specified deferral periods)
  • Recent exposure to blood or body fluids with risk of infection (with deferral periods)
  • Anemia or low blood cell counts outside specified limits
  • Recent blood or bone marrow donation (with required deferral periods)
  • Recent blood transfusion (deferral for 12 months)
  • Participation in incompatible research studies within last 6 weeks
  • Recent travel to malaria-endemic areas with specified deferral periods
  • Close contact with smallpox vaccination sites (with deferral periods)
