Effect of Smoking Amount on Salivary High Sensitive C-Reactive Protein Levels Using Different Centrifuge Settings in Healthy Adults

Researchers are studying how different amounts of smoking affect the levels of high sensitive C-reactive protein (hs-CRP) in saliva among healthy adults. The study focuses on non-smokers, light smokers (less than 10 cigarettes per day), and heavy smokers (more than 10 cigarettes per day) who are periodontally healthy. The goal is to understand how centrifugation speed and time impact the measurement of hs-CRP in saliva and to explore any connections between these values and smoking habits. Participants provide saliva samples collected while sitting upright with their heads tilted forward to allow saliva to pool naturally. The saliva is then processed by centrifugation at different speeds and durations. The clear portion of the sample is stored frozen until analysis. Clinical periodontal measurements such as probing depth, clinical attachment level, bleeding on probing, gingival index, and plaque index are also recorded. The levels of hs-CRP in saliva are measured using an enzyme-linked immunosorbent assay (ELISA). Participants undergo full-mouth periodontal exams and provide saliva samples during the study visit. Researchers analyze the hs-CRP levels in saliva collected under different centrifugation conditions and compare them across smoking groups. The primary outcome is the total amount of hs-CRP in saliva measured 24 hours after the clinical assessments. The study includes adults aged 18 to 30 years who are systemically and periodontally healthy, with no recent medication or periodontal treatment.
