Populations worldwide are aging, leading to an increased prevalence of Alzheimer's disease (AD) due to a lack of effective treatments. Researchers are investigating how the traditional Mediterranean diet, rich in fiber and polyphenols (PPs), may help prevent or slow cognitive decline and preserve healthy brain function. This study focuses on people aged 50 and older who have an increased risk of AD, exploring how polyphenol intake influences brain health through gut microbiome interactions and secondary bile acid metabolism. The study compares two groups: one receiving a polyphenol supplement called Juice Plus Essentials Berry Blend Capsules and the other receiving a micronutrient-matched placebo supplement. Participants will take these dietary supplements over a 12-month period. The study collects biological samples and cognitive assessments at three times: baseline (week 0), mid-study (month 6), and final visit (month 12) to examine changes during the intervention. Participants will collect stool samples at home and attend appointments for blood draws and cognitive testing at the three key time points. Researchers will measure levels of polyphenol-derived metabolites and microbiome changes in stool and blood, along with cognitive function focusing on executive function. Safety and adherence will be monitored throughout the year-long study to better understand the relationship between polyphenol intake, gut microbiota, and cognitive health.
