Researchers are evaluating the neural effects of psilocybin treatment in adults with moderate to severe Alcohol Use Disorder (AUD) in a Phase 2, double-blind, randomized, placebo-controlled clinical trial. The study aims to understand how psilocybin influences brain activity related to alcohol cues and negative emotions, alongside assessing clinical improvements in AUD symptoms using brain imaging, self-reports, and blood biomarkers. Participants will receive either psilocybin capsules (one 25 mg and one 5 mg capsule, total 30 mg) or matching placebo capsules. All participants will also attend three supportive therapy sessions led by a licensed clinician and a therapist from Silver Hill Hospital, ensuring consistent clinical care. The study includes baseline assessments and follow-up visits to monitor effects over time. During the trial, participants will undergo brain imaging (fMRI) to measure changes in brain activation and connectivity in response to alcohol and negative emotional cues. Researchers will also track alcohol use through self-reports and blood tests, along with questionnaires on quality of life, craving, mood, and impulsivity. The study monitors safety and adherence throughout, with a total follow-up period extending to 24 weeks.
