Study of microRNAs in Patients with Critical Limb Ischemia Compared to Controls with Abdominal Aortic Aneurysm

Researchers are investigating the role of microRNAs miRNAs in patients with critical limb ischemia CLI, a severe condition affecting blood flow to the lower limbs. The study involves 40 patients with varying severity of CLI, including those undergoing bypass surgery or major amputation, and compares their muscle tissue and blood samples to 40 control patients with abdominal aortic aneurysm AAA but no ischemia. The aim is to identify miRNAs and related gene targets that may be involved in CLI and to find potential disease biomarkers over a period of up to 10 years. The study collects muscle biopsies from different sites depending on patient groups for CLI patients undergoing bypass, biopsies are taken from the gastrocnemius muscle for those undergoing thigh or leg amputation, biopsies are collected from muscles near the amputation site as well as from the amputated limb. Control patients undergoing AAA surgery have biopsies taken from the sartorius muscle. Additionally, blood samples are collected from all participants before surgery to analyze circulating miRNAs. Participants will undergo tissue and blood sampling during their scheduled surgeries. Researchers will analyze miRNA expression and the RISC complex in collected muscle fragments and blood samples. The primary outcomes include identifying pathogenic miRNAs and disease biomarkers, with evaluations spanning from one to ten years. The study involves 80 participants in total and includes long-term follow-up to assess the significance of miRNAs in CLI.
