Role of Monocytes and Macrophages in Sarcoidosis and Tuberculosis to Study Granuloma Formation and Maintenance

Researchers are investigating sarcoidosis, a systemic inflammatory disease characterized by granuloma formation, which shares many features with tuberculosis. This study aims to understand the role of monocytes and macrophages in granuloma formation and maintenance by comparing patients with sarcoidosis to those with tuberculosis. The goal is to define a specific immune cell signature and explore factors contributing to chronic immune overactivation, potentially leading to new treatment approaches. Participants include patients diagnosed with sarcoidosis or tuberculosis. Blood samples will be collected to analyze immune cell signatures using various advanced laboratory methods such as flow cytometry, RNA sequencing, and epigenetic studies. These analyses will occur at diagnosis and during follow-up visits at 0, 6, and 12 months. The study also examines tissue samples to understand granuloma structure and immune cell interactions. During the study, participants will provide blood and tissue samples at defined intervals for detailed laboratory tests including cytokine levels, transcriptomic, and epigenomic analyses. Researchers will monitor immune cell activation over up to 12 months to identify differences between sarcoidosis and tuberculosis. This observational study involves no treatment intervention, focusing instead on understanding disease mechanisms and identifying potential diagnostic markers or therapeutic targets.
