This study is a single-arm, self-control, single-center study to investigate the molecular mechanism under exercise improving metabolism of patients with prediabetes. 40 overweight/obese patients with prediabetes will be recruited in this study and instructed to participate in an 8-week exercise program. We aim to investigate 1) the changes of clinical phenotypes such as body weight, body fat, laboratory values, and resting energy expenditure before and after exercise 2) the changes of peripheral blood mononuclear cell by single cell sequencing 3) search for potential biomarkers of exercise in improving metabolism through multi-omics integrated analysis of single-cell transcriptome, phenomics, metabolomics, etc.

Who Can Participate
Eligibility Criteria
You may qualify if you...
- Han Chinese
- Fasting blood glucose between 5.6 mmol/L and less than 7 mmol/L, and/or 2-hour postload glucose between 7.8 mmol/L and less than 11.1 mmol/L, and/or HbA1c between 5.7% and less than 6.5% without anti-hyperglycemic medication
- Voluntary participation with informed consent
You will not qualify if you...
- Participation in any weight-loss clinical study within the past 1 year
- Weight change of more than 10% of current weight in the past 1 year
- Use of certain drugs in the past 3 months such as hormone drugs, orlistat, metformin, GLP-1 receptor agonists, lipid-lowering agents, or liver protection drugs
- Diseases affecting glucose and lipid metabolism like hyperthyroidism, hypothyroidism, or Cushing syndrome
- Poorly controlled blood pressure: systolic ≥180 mmHg and/or diastolic ≥100 mmHg
- Serious heart, liver, kidney, or systemic diseases
- Skeletal and muscular diseases making exercise difficult
- Surgery within the past 1 year
- COVID-19 positive or recovered within 4 weeks
- Presence or history of eating disorders
- Presence of sleep disorders
- Employment involving disrupted circadian rhythms
- Pregnancy or breastfeeding
