Researchers are investigating the role of microbial diversity in pancreatic tissue across various pancreatic diseases, including pancreatic cancer, chronic pancreatitis, and other tumor types. The study aims to understand how different microbial flora in the pancreas relate to clinical and pathological features, potentially offering new insights for early diagnosis and treatment guidance. This observational research utilizes a large database of surgical patients from Ruijin Hospital to explore these relationships and mechanisms. The study involves analyzing paraffin-embedded pancreatic tissue samples collected from patients who underwent surgery between January 2005 and June 2022. High-throughput sequencing techniques, such as 16S rRNA gene sequencing and metagenomic analysis, are used to identify microbial populations within tumor cells, immune cells, and normal pancreatic cells. Researchers also apply immunohistochemical and immunofluorescence methods to detect microbial abundance and location in different pancreatic diseases. Participants have their clinical data reviewed, including symptoms, serological indexes, tumor stage, and prognosis, to correlate with microbiota findings. The study involves no intervention or treatment changes, relying solely on tissue samples and medical records. The primary outcome is the detailed microbial characterization of pancreatic tissues using 16S rRNA and metagenomic sequencing. The study will continue through July 2026, with no direct participant involvement beyond the use of stored surgical specimens and medical data.
