A Phase 2, Randomized, Double-Blind, Placebo-Controlled Study to Evaluate the Efficacy, Safety, and Tolerability of VIM0423 in Individuals With Isolated Dystonia

Researchers are evaluating VIM0423, a potential oral medication, for its ability to improve symptoms and quality of life in adults with isolated dystonia, a rare movement disorder causing abnormal muscle contractions and movements. This Phase 2, randomized, double-blind, placebo-controlled study aims to assess the safety, tolerability, and effectiveness of VIM0423 compared to a placebo in individuals with dystonia affecting two or more body parts. VIM0423 targets the chemical imbalance in brain regions believed to cause dystonia and is being developed to treat various forms of isolated dystonia beyond those currently approved for botulinum toxin treatment. Participants will be randomly assigned to receive either VIM0423 or a matching placebo, taking up to six pills each evening before bedtime for 16 weeks. The study includes groups with segmental/multifocal dystonia (two or more body parts affected) and generalized dystonia (more than two body parts affected), with about 120 participants expected to enroll. The study is conducted in phases including screening and baseline visits, dose titration and maintenance, dose taper, and a safety follow-up period. During the trial, participants will attend six in-person visits over up to 32 weeks. Researchers will assess changes in dystonia symptoms using video recordings and rating scales, along with clinical labs, EKGs, and monitoring for any adverse events. Participants will also complete self-assessments about the impact of dystonia on their daily lives. The primary outcome measure is the change in the Burke-Fahn-Marsden Dystonia Rating Scale from baseline to Week 14, with additional evaluations of clinical and patient impressions of change.
