What this Trial Is About
The goal of this study is to perform genetic sequencing on brain tumors from children, adolescents, and young adult patients who have been newly diagnosed with a high-grade glioma. This molecular profiling will decide if patients are eligible to participate in a subsequent treatment-based clinical trial based on the genetic alterations identified in their tumor.

Eligibility Criteria
You may qualify if you...
- Patients must be between 12 months and 39 years old at enrollment.
- Newly diagnosed with high-grade glioma (HGG), including DIPG, confirmed by biopsy or resection and local pathology review.
- DIPG diagnosis requires tumor centered in the pons with diffuse involvement of at least two-thirds of the pons and histopathology consistent with WHO grade 2-4 diffuse glioma.
- Other tumors must be WHO grade 3-4 gliomas.
- No specific disease status required; patients with measurable or non-measurable, metastatic, multifocal, or spinal tumors are eligible.
- Prior surgery, radiation, dexamethasone, and temozolomide with radiation are allowed; prior bevacizumab use allowed with arm-specific washout.
- Radiation must start within 42 days of surgery or biopsy; strongly recommended within 31 days.
- Tumor sample plus normal tissue must be submitted for molecular profiling, or previous profiling results must be provided.
- Written informed consent (and assent when appropriate) must be obtained.
- Enrollment timing varies by screening option but generally between diagnosis and up to 31 days post definitive surgery or post radiation depending on option.
You will not qualify if you...
- Tumors that do not meet the specified high-grade glioma or DIPG diagnoses.
