Researchers are evaluating whether the drugs retatrutide and tirzepatide can prevent major adverse liver outcomes (MALO) in adults with metabolic dysfunction-associated steatotic liver disease (MASLD) who are at high risk. This Phase 3 trial enrolls about 4,500 adults with MASLD identified by non-invasive tests indicating an increased likelihood of developing serious liver problems. The study aims to understand how these treatments might affect liver health over time compared to a placebo. Participants will be randomly assigned to receive either retatrutide, tirzepatide, or a placebo, all given by subcutaneous injection. The study will last approximately 224 weeks, during which participants may attend 25 to 30 clinic visits for monitoring and assessment. After the main study, eligible participants can join an optional 2-year extension where all will receive either retatrutide or tirzepatide regardless of their original group. Throughout the trial, participants’ liver function and disease progression will be closely monitored through various health assessments. Researchers will track the time to the first major adverse liver event as the main outcome. Safety and health status will be evaluated regularly during clinic visits, ensuring thorough observation over the long study period.

Researchers are investigating the effectiveness, safety, and tolerability of combining baxdrostat with dapagliflozin compared to dapagliflozin alone in people with chronic kidney disease (CKD) and high blood pressure. This Phase III, international, multicenter, double-blind, placebo-controlled study aims to see if this combination reduces risks such as significant kidney function decline, kidney failure, heart failure events, or cardiovascular death. The study includes a 4-week run-in period where participants not previously treated with SGLT2 inhibitors receive dapagliflozin alone. After this, participants are randomly assigned to receive either baxdrostat plus dapagliflozin or placebo plus dapagliflozin in a double-blinded manner. Study visits occur frequently initially (at 2, 4, 8, 16, 34, and 52 weeks after randomization) and then approximately every 4 months. If participants stop the blinded treatment early, they continue dapagliflozin alone unless specific criteria require its discontinuation. Participants will undergo regular assessments including blood pressure monitoring and laboratory tests related to kidney function and cardiovascular health. The primary outcome measures the reduction in risk of major kidney and heart events over up to 37 months. Even if participants stop the study treatment, they will continue follow-up visits and data collection to ensure comprehensive safety and efficacy evaluation throughout the study duration.

Researchers are evaluating AZD0780, an oral PCSK9 inhibitor, in a phase 3, randomized, placebo-controlled study to see if it can reduce the risk of major adverse cardiovascular events (MACE-PLUS) in adults with established atherosclerotic cardiovascular disease (ASCVD) or those at high risk for a first ASCVD event. The study compares AZD0780 to a placebo and monitors participants from randomization until the primary analysis censoring date, followed by a final study closure visit. Participants will be randomly assigned to receive either oral AZD0780 or an oral placebo once daily. The treatment period lasts until the primary analysis censoring date, after which a study closure visit will occur. The study is event-driven and designed to assess the time to the first major cardiovascular event during treatment. During the study, participants will be closely monitored with various assessments to evaluate cardiovascular outcomes and safety over approximately 54 months. Researchers are evaluating the safety and effects of different doses of a new medicine called NNC0519-0130 on kidney function in adults with chronic kidney disease, some of whom may also have type 2 diabetes, and who are living with overweight or obesity. The study compares NNC0519-0130 with semaglutide, an existing medicine, and a placebo, which is a "dummy" treatment. This is a Phase 2 proof-of-concept and dose-finding study aimed at understanding how these treatments may reduce kidney damage. Participants will be randomly assigned to one of three groups receiving either NNC0519-0130, semaglutide, or placebo. All treatments are given by subcutaneous injection once weekly. The study treatment phase lasts up to 36 weeks, with assessments at weeks 12, 24, and 36 to monitor changes in kidney damage by measuring the urinary albumin-to-creatinine ratio. The overall study duration can be up to 43 weeks. During the study, participants will be regularly monitored through laboratory tests and clinical evaluations to assess kidney function and safety. Researchers will measure changes from the start of the study in the urinary albumin-to-creatinine ratio at multiple time points. Participants will also need to have stable doses of certain blood pressure medications before joining. Safety and treatment effects will be assessed throughout the study period.

Researchers are evaluating the persistence of the immune response and the safety and immune response after revaccination with the adjuvanted RSVPreF3 vaccine in adults aged 18 and older who have received lung or kidney transplants. This phase 2b extension study focuses on individuals who previously received one or two doses of this vaccine in an earlier study and are now receiving an additional dose to assess continued protection and safety. Participants who received either one or two doses of the adjuvanted RSVPreF3 vaccine in the prior RSV OA=ADJ-023 study will receive a single intramuscular dose of the same vaccine at the start of this study (Day 1). They will be grouped based on their initial dosing for immune response analyses but combined for safety and demographic evaluations. Throughout the study, participants will have visits on Day 1, Day 31, and Day 180 to measure immune responses against RSV-A and RSV-B through neutralizing antibody levels. Safety and immune system responses will be monitored, including detailed evaluations of antibody levels at each visit to understand vaccine persistence and revaccination effects over time.

Researchers are evaluating the combination of ASP-1929 photoimmunotherapy (PIT) with pembrolizumab compared to pembrolizumab alone or pembrolizumab plus chemotherapy in treating patients with locoregional recurrent squamous cell carcinoma of the head and neck (HNSCC) without distant metastases. This phase 3 multicenter, randomized study aims to determine if adding ASP-1929 PIT improves overall survival and to assess the safety of this combined treatment. Participants will be randomly assigned in a 2:2:1 ratio to receive either ASP-1929 PIT at doses of 320 mg/m² or 640 mg/m² combined with pembrolizumab, or standard of care treatment which includes pembrolizumab alone or pembrolizumab plus chemotherapy such as carboplatin, cisplatin, 5-fluorouracil, paclitaxel, or docetaxel based on the physician's choice. Treatment with ASP-1929 involves an intravenous infusion followed by illumination with light within 24 hours to activate the therapy. Pembrolizumab is given intravenously every 3 or 6 weeks. Chemotherapy, if chosen, is given intravenously every 3 weeks for up to 6 cycles. Throughout the study, participants will be monitored for up to approximately 48 months to evaluate their overall survival. Researchers will perform assessments including tumor measurements and safety evaluations. An interim analysis will guide potential adjustments in dosing. The study includes careful monitoring of organ function, performance status, and adverse events to ensure participants' safety during and after treatment.

This research aims to evaluate the safety and effectiveness of the combination of avutometinib and defactinib compared to standard treatments chosen by investigators in women with recurrent low-grade serous ovarian cancer (LGSOC) who have experienced disease progression after prior platinum-based therapy. Both avutometinib and defactinib are investigational kinase inhibitors designed to block cancer cell growth. The study will also assess overall survival, other measures of treatment effectiveness, safety, and quality of life impacts. Participants will be randomly assigned to receive either the combination of oral avutometinib and defactinib or one of four standard treatments recommended for recurrent LGSOC: pegylated liposomal doxorubicin and paclitaxel (both given intravenously), or the oral drugs letrozole or anastrozole. Patients treated with standard therapies who experience disease progression may be eligible to switch to the investigational combination. The study is open-label and conducted internationally by specialists in gynecological cancer. Throughout the study, participants will have regular follow-up visits including scans and tests to measure disease progression, with a primary focus on progression-free survival up to 24 months. Researchers will monitor safety, side effects, and overall survival while collecting information on quality of life and symptoms. The study involves ongoing assessments of treatment effects and participant health until the study concludes or disease progression occurs.
