Researchers are studying the digestive system and its function in adults aged 18 to 65 who have previously undergone bariatric surgery, such as Roux-en-Y gastric bypass or sleeve gastrectomy, at least one year ago, as well as in adults living with obesity. The study aims to understand how digestion and gastrointestinal physiology work after these surgeries, focusing on enzyme activity, bile acid levels, pH, and transit times within the digestive tract. The trial involves two main study visits. During the first visit, fluids from the stomach and small intestine will be collected using a naso-intestinal catheter. In the second visit, participants will swallow a special ingestible capsule that continuously measures various physiological conditions throughout the entire gastrointestinal tract. These interventions help researchers evaluate enzyme activities and digestive parameters across different regions of the digestive system. Participants will undergo these procedures to provide samples and physiological data that researchers will analyze to measure activities of several enzymes including gastric lipase, pepsin, trypsin, chymotrypsin, alpha-amylase, and pancreatic lipase within 2 hours. The study will also monitor bile acid concentrations, pH, and gastrointestinal transit time. Throughout the trial, participant safety and adherence will be closely observed, and the total study duration includes both visits with detailed physiological assessments.
