Researchers are evaluating the safety and effectiveness of Armour Thyroid compared to synthetic T4 treatment in adults with primary hypothyroidism who are currently stable on synthetic T4. The study focuses on assessing how well patients respond to dose conversion from synthetic T4 therapy to Armour Thyroid. This trial is conducted as a Phase 2/3 multicenter, double-blind, randomized, active-controlled study. Participants receive either Armour Thyroid in oral capsule or tablet form or synthetic T4 capsules. They must have been on a stable dose of synthetic T4 for at least 12 months before screening, with a dose of at least 25 mcg daily. The study compares both treatments over time to evaluate efficacy and safety in maintaining thyroid function. During the study, researchers monitor thyroid-stimulating hormone (TSH) levels to measure treatment response at week 55. They also track any adverse events related to the treatments for up to approximately 90 weeks. Participants undergo regular assessments to ensure safety and effectiveness throughout the study period.

The purpose of the study is to evaluate whether ibuzatrelvir is effective and safe in adults and adolescents with COVID-19 who do not need to be in the hospital but who are at high risk for progression to severe disease. Eligible participants will be randomly assigned (by chance) to receive ibuzatrelvir or matching placebo orally for 5 days. Co-administration of locally available standard of care is allowed. The total duration of the study is around 6 months.

Researchers are studying adults with hypertriglyceridemia (HTG) and severe hypertriglyceridemia (SHTG) who have completed previous related studies. The main goal is to evaluate the long-term safety and effectiveness of plozasiran, a drug given by injection, in these adults. Participants must meet specific health criteria, including controlled blood sugar levels and prior study completion, to join this open-label phase 3 extension trial. Eligible participants will receive plozasiran injections under the skin about every three months for two years. They will be advised to continue a low-fat diet throughout the study. This study includes adults from various countries who have met all previous study requirements or were prevented from randomization to avoid over-enrollment but still meet eligibility. Special criteria apply for some participants from earlier studies regarding their triglyceride levels and history of pancreatitis. During the study, participants will be monitored for any treatment-related side effects from the first dose through month 24. Researchers will assess safety by tracking adverse events and other health measures. Participants will also be counseled on medication adherence and diet, with ongoing evaluations to ensure their well-being throughout the two-year treatment period.

Researchers are evaluating surgical and minimally invasive treatments for lumbar spinal stenosis (LSS) by comparing Medicare patients who received the MILD procedure against those who had interspinous process decompression (IPD). The study focuses on outcomes such as the rate of harms related to the initial procedure and the frequency of additional surgical or minimally invasive interventions within 24 months after treatment. Enrollment includes patients treated from January 1, 2017, onward, with continuation until the sponsor decides to stop. The MILD procedure involves percutaneous image-guided lumbar decompression, performed under fluoroscopy through a dorsal approach to partially remove tissue and bone at the affected spinal level. The control group receives the IPD procedure for LSS. Both groups are monitored for a 24-month period post-index procedure using Medicare claims data to track reoperations and any harms. Participants contribute data through Medicare claims without needing prior enrollment or consent, as the study is exempt from IRB oversight. Researchers collect and analyze information on procedure-related harms and subsequent interventions over two years. This approach allows evaluation of long-term safety and effectiveness outcomes for patients treated with either MILD or IPD.

Researchers are evaluating the safety and lasting effects of AR-14034 Sustained Release (SR) implant in people with neovascular age-related macular degeneration (nAMD). This study has two stages, starting with a dose-escalation phase to assess safety and preliminary effects, followed by a randomized comparison with an approved treatment called aflibercept. The trial is designed as a Phase 1/2 study to better understand how well AR-14034 SR works and how long its benefits last compared to standard therapy. In Stage 1, about 10 participants will receive one of two dose levels of AR-14034 SR via an intravitreal implant and be followed for 48 weeks to monitor safety and early treatment response. In Stage 2, approximately 130 participants will be randomly assigned to one of three groups: one injection of AR-14034 SR, two injections of AR-14034 SR, or aflibercept injections as currently labeled. This stage lasts 56 weeks, followed by a 16-week open-label extension phase where participants continue to be observed. Participants will attend multiple visits throughout the study to assess visual acuity using standard eye charts and monitor safety. Researchers will collect data at baseline, Week 44, and Week 48 to measure changes in vision. Additional assessments include eye exams and evaluations for any side effects during and after treatment. The total duration for participants ranges from 48 to 72 weeks depending on the stage and extension phase involved.
