Researchers are evaluating efruxifermin EFX in adults with non-cirrhotic nonalcoholic steatohepatitis NASH or metabolic dysfunction-associated steatohepatitis MASH who have liver fibrosis stage 2 or 3. This Phase 3, multi-center, randomized, double-blind, placebo-controlled study aims to assess the safety and efficacy of EFX compared with placebo. The trial includes about 1,650 participants divided into two cohorts based on liver biopsy characteristics and fibrosis stage. Participants will be randomly assigned to one of three groups EFX 28 mg, EFX 50 mg, or placebo, each given as a weekly subcutaneous injection. Cohort 1 will be evaluated over 52 weeks for histologic efficacy endpoints, while Cohort 2 will have assessments over 96 weeks. After these periods, participants may continue long-term treatment and clinical follow-up for up to approximately 240 weeks total. A follow-up visit will occur about 30 days after the last dose. During the study, participants will undergo liver biopsies, blood tests, and non-invasive assessments such as FibroScan and Enhanced Liver Fibrosis ELF score to monitor liver health and fibrosis. Researchers will track liver-related clinical outcomes, including liver events and survival, as well as safety and tolerability of the treatment. Participants who stop the study drug may still continue with scheduled assessments to support long-term safety and efficacy evaluations.

Researchers are studying obefazimod to evaluate its effectiveness and safety as a treatment for adults with moderately to severely active Crohns disease who have not responded well or cannot tolerate conventional or advanced therapies. This Phase 2b trial compares obefazimod with a placebo to see if it can help control symptoms and improve disease activity. The study also aims to assess the long-term safety and tolerability of obefazimod during an extension period. The study includes three treatment phases a 12-week induction phase, a 40-week maintenance phase, and a 48-week extension phase. The trial investigates the prevention of deep infections in patients with severe open tibia fractures. It compares outcomes between those treated with a new antibiotic coated intramedullary nail and those receiving the traditional standard of care intramedullary nail. The study focuses on reducing infections that can lead to disability, rehospitalization, and in severe cases, amputation, especially among active duty military personnel. Participants are randomly assigned to one of two groups one receives an intramedullary nail coated with a mixture of vancomycin and gentamicin antibiotics, while the other receives a standard uncoated intramedullary nail. The antibiotic coating is applied using a cement mixture during the definitive fixation surgery. The trial follows patients for one year after treatment. During the 12-month follow-up, participants undergo evaluations at baseline, 6 weeks, 3 months, 6 months, and 12 months. Researchers are studying rapid reinitiation of antiretroviral therapy ART with bictegraviremtricitabinetenofovir alafenamide BFTAF in people living with HIV who have interrupted their treatment for at least 12 weeks. The trial aims to evaluate the safety and effectiveness of starting BFTAF on the same day participants return to care. This approach addresses challenges that cause delays in restarting treatment, such as waiting for lab results and multiple medical visits, which may discourage people from resuming care. Participants will receive open-label BFTAF tablets once daily for either 24 or 48 weeks, depending on their viral load at baseline. Those with HIV-1 RNA levels above 50 copiesmL will be treated for 48 weeks, while those with suppressed viral loads below 50 copiesmL will receive 24 weeks of treatment. Treatment begins immediately during the initial visit without waiting for baseline lab results, allowing for rapid reinitiation. The study includes two cohorts based on viral load and optional interviews to understand reasons for treatment interruption and reinitiation. During the study, participants will be monitored through clinical assessments, viral load measurements, CD4 cell counts, safety evaluations, and adherence tracking. Patient-reported outcomes will also be collected to capture experiences with the therapy and treatment satisfaction. Follow-up lasts up to 48 weeks for viremic participants and 24 weeks for suppressed participants, with interviews conducted by teleconference at weeks 4 and 24. The research team will measure how many participants achieve viral suppression and remain engaged in care.
