Researchers are studying people with idiopathic pulmonary fibrosis IPF or progressive pulmonary fibrosis PPF who have previously taken nerandomilast in another study. The aim is to assess how well patients tolerate long-term treatment with nerandomilast and to evaluate whether it improves lung function and delays worsening symptoms, hospital visits, or death. This open-label extension trial is sponsored by Boehringer Ingelheim and focuses on treatment over an extended period. Participants take nerandomilast tablets for up to 1 year and 10 months while continuing their usual pulmonary fibrosis treatments. The study involves a single treatment group receiving the drug, and no placebo or comparison groups. Regular visits with doctors help monitor health and collect data during this extended treatment phase. Throughout the study, participants undergo regular lung function tests and health assessments to track any adverse events and changes in lung capacity. The main outcome measured is the occurrence of any adverse events for up to about 99 weeks. Secondary outcomes include changes in forced vital capacity and time to worsening of disease symptoms or hospitalization. The study includes ongoing safety monitoring with a total participation time of up to nearly two years.

Researchers are evaluating the efficacy and safety of elecoglipron, an oral tablet taken once daily, for weight management in adults with obesity or overweight. This Phase III global, randomized, double-blind, placebo-controlled trial includes two independent pivotal studies one in adults without type 2 diabetes T2DM and the other in adults with T2DM, all having at least one weight-related health condition. The goal is to understand how elecoglipron compares to placebo when combined with diet and exercise. Participants will be randomly assigned to receive either one of two doses of elecoglipron or a matching placebo daily. Study 1 involves about 3000 adults living with obesity or overweight without T2DM, while Study 2 involves about 1500 adults with obesity or overweight and T2DM. Both studies last 72 weeks, during which changes in body weight and other health measures will be monitored. During the trial, participants will undergo regular health assessments including measurements of body weight, waist circumference, blood sugar control, blood pressure, and other related health indicators. Researchers will track percent change in body weight from baseline at 72 weeks as the primary outcome. Participants will be monitored closely throughout the study to assess safety and effectiveness of the treatment in managing weight and associated health conditions.

Researchers are evaluating zelquistinel, a drug aimed at reducing symptoms of major depressive disorder in adults aged 18 to 64 years. This Phase 2 clinical trial compares the effects and safety of zelquistinel to a placebo in participants diagnosed with major depressive disorder. The study will focus on changes in depression severity and monitor any medical issues that arise during treatment. Participants will take one tablet of either zelquistinel or placebo once a week for six weeks. Researchers are evaluating AZD4512, an antibody-drug conjugate targeting CD22, for its safety and effectiveness in adults with relapsed or refractory B-cell Non-Hodgkin Lymphoma B-NHL. This Phase III open-label global study explores AZD4512 alone or combined with other anticancer agents, focusing on participants who have previously undergone at least two lines of therapy. The trial also aims to study pharmacokinetics, pharmacodynamics, and immunogenicity of AZD4512 in this population. The study includes a dose escalation phase and a pharmacodynamicsafety backfill phase to determine the maximum tolerated dose or optimal biological dose of AZD4512 monotherapy. Participants receive AZD4512 through intravenous infusion, and additional modules may be added to study specific B-NHL subtypes or combination treatments. This modular design allows for assessment of AZD4512 alone or with other anticancer agents across various B-NHL subgroups. Participants will be monitored for dose-limiting toxicities, adverse events, and changes in vital signs or laboratory tests from the first dose through 30 days after the last dose. Researchers are evaluating the effects of ensifentrine inhalation suspension compared to a placebo in adults with non-cystic fibrosis bronchiectasis NCFBE. This study aims to understand how ensifentrine, added to standard care, impacts lung flare-ups, symptoms, and quality of life. It is a randomized, double-blind, placebo-controlled Phase II trial designed to assess both efficacy and safety over up to 52 weeks. Participants will be randomly assigned to receive either 3 mg of nebulized ensifentrine or a placebo, both delivered twice daily using a standard jet nebulizer. The treatment period may last up to 52 weeks, although the study may end earlier once the last participant completes 24 weeks. Neither participants nor study staff will know which treatment is administered during the study. During the study, participants will be monitored regularly, including assessments of lung function, respiratory symptoms, quality of life, and tracking of any pulmonary exacerbations. Measurements will be taken at baseline and at various time points up to 52 weeks. Safety will be evaluated through adverse event tracking. The primary outcome focuses on the rate of pulmonary exacerbations per year, with additional assessments of symptom changes and lung function.

Researchers are evaluating the effects of a triple therapy inhaler combining budesonide, glycopyrronium, and formoterol fumarate BGF MDI 32014.49.6 g compared to a dual therapy inhaler with glycopyrronium and formoterol fumarate GFF MDI 14.49.6 g on heart and lung outcomes in adults with Chronic Obstructive Pulmonary Disease COPD who have a higher risk for heart and lung events. This Phase III study is randomized, double-blind, and conducted at multiple centers, focusing on participants with COPD and elevated cardiopulmonary risk. Participants will receive either the triple therapy inhaler or the dual therapy inhaler, both administered twice daily. The study compares these two inhalers over a period of up to three years, monitoring for serious cardiac or COPD events. The trial includes careful evaluation of various heart and lung-related health events during this period. During the study, participants will be closely monitored through regular visits, assessments, and tests to measure lung function, heart events, and COPD exacerbations. This trial studies adults aged 55 to 90 who have psychosis linked to Alzheimers Disease. It is a Phase 3, 38-week, randomized, double-blind, placebo-controlled outpatient study. Its main goal is to assess how well KarXT capsules prevent relapse of psychosis compared to placebo. Additional goals include evaluating time to treatment discontinuation or relapse, and monitoring safety and tolerability. Participants receive either KarXT capsules at various doses or placebo capsules. The study involves a randomized assignment and is conducted under quadruple masking. The treatment period lasts 38 weeks during which KarXT or placebo is taken three times daily. Assessments continue up to approximately 42 weeks to monitor adverse events and other safety measures. Throughout the study, participants attend outpatient visits for evaluations including cognitive tests, assessments of psychosis severity, caregiver reports, lab tests, vital signs, and safety monitoring. Researchers are evaluating the effectiveness and safety of a new drug called GSK3862995B in adults with bronchiectasis, a chronic lung disease. This Phase 2 study also examines how the body processes the drug and checks for any immune reactions. Participants will be randomly assigned to receive one of two doses of GSK3862995B or a placebo to compare their effects on bronchiectasis. Participants will receive either Dose Level 1 or Dose Level 2 of GSK3862995B, or a matching placebo. The study uses a double-blind, randomized design, meaning neither participants nor researchers know who gets which treatment. The treatment period lasts up to 48 weeks, during which participants receive repeated doses. The study also includes assessments up to 72 weeks to monitor safety and immune responses. Researchers are studying two surgical procedures to reduce the risk of ovarian cancer in women with BRCA1 genetic mutations. This trial compares bilateral salpingectomy, which removes only the fallopian tubes, with bilateral salpingo-oophorectomy, which removes both fallopian tubes and ovaries. The goal is to find out if removing just the fallopian tubes with delayed ovary removal is nearly as effective as removing both from the start. Participants choose between two groups one undergoes bilateral salpingectomy with the option of later ovary removal, and the other undergoes bilateral salpingo-oophorectomy. Both groups have imaging tests like pelvic ultrasounds or pelvic MRIs during screening and provide blood samples throughout the study. Follow-up visits occur at multiple time points, including 10 to 60 days, 6 months, 12 months, 24 months, and then yearly for up to 20 years. During the study, researchers track if ovarian or related cancers develop and assess symptoms related to estrogen loss, quality of life, cancer-related distress, sexual function, menopausal symptoms, medical decision making, and any adverse events. Various questionnaires and imaging tests support these evaluations. Long-term safety and cancer risk reduction are monitored for up to two decades after surgery.

This research is focused on adults aged 40 and older who have idiopathic pulmonary fibrosis IPF, a lung condition characterized by scarring and reduced lung function. The study aims to evaluate whether the medicine BI 765423 can improve lung capacity compared to a placebo. Participants must have certain lung function levels and evidence of lung fibrosis confirmed by imaging to join the study. Participants are randomly assigned to one of two groups one receives BI 765423 through an intravenous infusion every four weeks, and the other receives a placebo that looks the same but contains no active medicine. The study lasts up to 11 months, during which participants may continue their standard IPF treatments. The main treatment period focuses on observing changes in lung function over 3 to 6 months.
