Researchers are investigating how gallotannin-rich mangoes and the probiotic Lactiplantibacillus pentosus may affect inflammatory markers and the gut microbiome in obese adults. The study aims to understand whether these treatments, alone or combined, influence gut bacteria linked to obesity and inflammation, and how genetic differences may impact gallotannin metabolism. This pilot study compares obese participants receiving different treatments to a lean control group to explore potential anti-inflammatory effects and microbial changes. Participants will be divided into four groups: mango juice only, mango plus L. pentosus, L. pentosus only, and a lean control group with no treatment added. The mango used is high in polyphenols called gallotannins, which gut microbes can break down into beneficial metabolites. The probiotic L. pentosus is studied for its gut health and immune benefits. Each participant will provide biological samples including saliva, blood, stool, and buccal cells, but will visit the collection site only once during the study week. Throughout the study, researchers will analyze inflammatory biomarkers, gut microbial composition using DNA sequencing, and levels of gallotannin metabolites using advanced chemical analysis. They will also perform genetic testing to assess variations affecting metabolism. The primary outcome measures include changes in inflammatory markers and gut microbiome diversity in vitro after 12 hours of treatment. This comprehensive approach will help evaluate the potential synergistic effects of mango polyphenols and probiotics on obesity-related inflammation and gut health.
