Study of Lung and Intestinal Microbiome and Host Factors in Adults With Severe Pneumonia

Researchers are conducting a prospective, multicenter cohort study to investigate severe pneumonia. The study aims to understand the relationship between the lung and intestinal microbiomes, host susceptibility, and patient prognosis. This research addresses gaps from previous small, single-center studies by including a larger sample size and diverse patient data. It focuses on severe pneumonia cases caused by various pathogens admitted to intensive care units ICUs. The sponsor of this study is the First Affiliated Hospital of Zhejiang University. The study collects clinical samples including sputum, alveolar lavage fluid, feces, and whole blood from severe pneumonia patients admitted to ICUs between 2023 and 2025. Patients are included based on specific criteria such as receiving mechanical ventilation, high flow oxygen therapy, or oxygen treatment with partial respiration masks meeting defined oxygen levels. Clinical metagenomics is used to evaluate pathogens present in these samples, helping to identify changes in lung and intestinal microbiomes and their connection to disease progression. Participants provide samples and clinical data that will be analyzed to measure the correlation between the lung microbiome and disease prognosis over 60 days. The study observes patients health status and disease progression risks while monitoring host susceptibility. This observational study involves no intervention but collects detailed biological and clinical information to gain insights into severe pneumonia. Participants will be followed during their ICU stay and throughout the 60-day observation period.
