A Study of Vedolizumab in Children and Teenagers With Moderate to Severe Crohn's Disease (CD)

What this Trial Is About
Vedolizumab is a medicine that helps to reduce inflammation and pain in the digestive system. In this study, children and teenagers with moderate to severe Crohn's disease will be treated with vedolizumab. The main aim of the study is to check if participants achieve remission after treatment with the vedolizumab. Remission means symptoms improve or disappear and an endoscopy shows no signs of inflammation. Participants will receive 3 infusions of vedolizumab over 6 weeks. Then, those who have a clinical response will receive either a high dose or low dose of vedolizumab once every 8 weeks. They will receive the same dose every time.

Who Can Participate
Eligibility Criteria
You may qualify if you...
- Child has moderate to severe Crohn's disease unresponsive or intolerant to current standard treatments
- Child weighs at least 10 kg at screening and enrollment
- Crohn's disease diagnosis made at least 1 month before screening
- Pediatric Crohn's Disease Activity Index (PCDAI) score over 30 and Simple Endoscopic Score for Crohn's Disease (SES-CD) over 6 (or over 4 if disease is limited to terminal ileum) at screening
- Failed, lost response to, or intolerant of corticosteroids, immunomodulators, or TNF-alpha antagonist therapy
- If extensive or left-sided colitis present for many years, child must have a recent negative colonoscopy within 12 months
- Vaccinations up to date based on country guidelines
You will not qualify if you...
- Received investigational biologic within 60 days or 5 half-lives before screening, or approved biologic/biosimilar within 2 weeks before study drug or during screening
- Active cerebral or meningeal disease, history of progressive multifocal leukoencephalopathy (PML) or major neurological disorders
- Clinically significant infection within 30 days prior to first dose
- Received any live vaccinations within 30 days prior to first dose
- Currently require or expected to require surgical intervention for Crohn's disease during study
- History of subtotal or total colectomy or certain intestinal surgeries/stomas
- Diagnosis of indeterminate colitis
- Signs of monogenic very early-onset inflammatory bowel disease
- Active or latent tuberculosis, positive TB test within 30 days of screening
- Positive hepatitis B surface antigen or core antibody without confirmed absence of HBV DNA
- Chronic hepatitis C infection
- Identified congenital or acquired immunodeficiency
- Evidence of dysplasia or history of malignancy except certain skin or cervical cancers
- Positive stool tests for parasites, culture, or Clostridioides difficile at screening
