What this Trial Is About
Researchers are investigating the genetic makeup of advanced solid tumors and primary central nervous system CNS cancers to understand how comprehensive genomic profiling CGP from tumor tissues or circulating tumor DNA ctDNA can guide treatment decisions. This observational study aims to describe the prevalence of specific genetic alterations and evaluate how molecular tumor board MTB recommendations impact treatment choices across different tumor types, with a focus on breast and lung cancers. Participants will undergo either tissue-based CGP using Roche AVENIO Tumor Tissue CGP kits or liquid biopsy testing with FoundationOne Liquid CDx when tissue is unavailable. More than 500 patients with available tissue samples will have their tumors profiled centrally at UZ Leuven, while about 50 patients without tissue will receive liquid biopsy testing. The sequencing data will be further analyzed by collaborating laboratories. Tumor cohorts for breast and lung cancer will be prioritized, with limits on enrollment for other tumor types. The MTB will review genomic data to provide treatment recommendations. During the study, participants will provide archived tissue samples no older than two years or undergo liquid biopsy. Researchers will monitor the distribution of actionable genetic alterations, the number of participants receiving and following MTB treatment recommendations, turnaround times for testing, and technical success rates. The study will also assess the impact on patient referrals for clinical trials. Participation involves data collection and analysis over an average period of one year, with safety and treatment uptake monitored throughout.
