What this Trial Is About
This research will establish and continuously improve the FAD research network in conjunction with multi-center institutions nationwide. By collecting information on the family's demography, genetics, neuropsychology, neuroimaging, biomarkers and other information, we can understand the current FAD population in China, clarify the genetic characteristics, pathogenesis, disease characteristics and diagnosis and treatment status of AD in China; which will lay the foundation for ameliorating clinical diagnosis and treatment, establishing a Chinese FAD clinical database and an international cooperative research platform. 1. To set up a multi-center, nationwide FAD research network and database platform in China 2. To clarify the epidemiological characteristics of FAD in China. 3. To clarify the genetic characteristics of FAD in China. 4. To clarify the clinical characteristics and disease development laws of FAD. 5. To discover and verify the early diagnosis biomarkers of AD. 6. To establish a genetic counseling model.

Who Can Participate
Eligibility Criteria
You may qualify if you...
- Written informed consent obtained from the participant or legal guardian before any study procedures
- At least two first-degree relatives in the family have Alzheimer's disease, with at least three patients across two generations
- At least one family member has normal cognitive function and is older than the average age of onset in the family
- Pedigrees carry familial Alzheimer's disease pathogenic genes (APP, PSEN1, PSEN2)
- Participants are over 18 years old
- Participant is cognitively normal or has dementia but is not bedridden
- Able to provide two reliable informants for clinical information
- Dementia diagnosed according to DSM-IV-R criteria
- Alzheimer's diagnosis based on NINCDS-ADRDA or NIA-AA criteria
- Mild cognitive impairment diagnosed according to Petersen criteria and classification by Lopez et al.
- Normal control group participants are 18 years or older with normal MMSE and MoCA scores adjusted for education levels
You will not qualify if you...
- Dementia caused by other factors such as depression, psychiatric illnesses, thyroid dysfunction, encephalitis, multiple sclerosis, brain trauma, brain tumors, infections, or other types of dementia like vascular dementia, frontotemporal dementia, dementia with Lewy bodies, or Parkinson's dementia
- MRI and lab tests do not support Alzheimer's diagnosis
- Severe circulatory, respiratory, urinary, digestive, hematopoietic diseases or cancer
- Severe psychiatric illness or severe dementia that prevents completing assessments
- History of alcoholism or drug abuse
- Pregnant or lactating women
- No reliable informant available
- Lumbar puncture contraindications such as coagulation disorders, recent lumbar surgery, or chronic CSF leakage
- MRI contraindications including metal implants, metallic foreign bodies in the eye, or brain aneurysm clips
- Normal controls excluded for abnormal MMSE or MoCA, history of brain injury or infarction, neurological diseases, systemic diseases causing cognitive impairment, mental retardation, inability to undergo cognitive exams, MRI contraindications, refusal of blood draw or consent
