Researchers are evaluating the effects of pelacarsen (TQJ230), given as a monthly injection under the skin, in people with mild to moderate calcific aortic valve stenosis. This study aims to see if pelacarsen can safely slow the progression of this heart valve condition compared to a placebo. The trial is a phase 2, randomized, double-blind, placebo-controlled study conducted at multiple centers. Participants will receive either pelacarsen 80 mg or a matching placebo once a month. Before starting the treatment, they must have elevated lipoprotein(a) levels and be optimally treated for existing cardiovascular risk factors. The study focuses on those aged 50 to under 80 years with mild or moderate calcific aortic valve stenosis. During the 36 months of participation, researchers will monitor changes in peak aortic jet velocity and aortic valve calcium score to assess disease progression. Safety, tolerability, and the impact of the treatment will be evaluated. Participants will undergo regular assessments, including laboratory tests and clinical evaluations, to track heart valve condition and overall health throughout the study.

Researchers are evaluating a new combined approach using multifocal non-invasive brain stimulation along with brain recording and cognitive training to treat cognitive and mood symptoms in patients with mild cognitive impairment (MCI) and early dementia. This study explores whether this integrated protocol can reduce these symptoms and whether factors like education and cognitive reserve influence the treatment's effectiveness. It also aims to identify which patients respond best to the brain stimulation by combining it with brain activity monitoring. The treatment involves two cycles of either real or placebo multifocal transcranial direct current stimulation (tDCS) applied with 2.0 mA intensity across eight electrodes placed over the left frontal and temporal brain regions. Each real stimulation session lasts 20 minutes, while placebo stimulation mimics the sensation but lasts only 30 seconds. Both cycles include traditional cognitive therapy with paper-and-pencil and computer exercises targeting memory, attention, and executive functions, performed twice weekly over two months. Participants will undergo neuropsychological tests before starting treatment and after each cycle to assess cognitive and mood changes. Tests include the Montreal Cognitive Assessment, Frontal Assessment Battery, Free and Cued Selective Reminding Test, Stroop Test, and Geriatric Depression Scale, all administered at baseline, week 4, and week 8. Caregivers will provide information about daily living activities. The study closely monitors responses to treatment to better understand who benefits from this combined intervention and how mood and cognition are affected over time.

Researchers are evaluating AZD0780, an oral PCSK9 inhibitor, in a phase 3, randomized, placebo-controlled study to see if it can reduce the risk of major adverse cardiovascular events (MACE-PLUS) in adults with established atherosclerotic cardiovascular disease (ASCVD) or those at high risk for a first ASCVD event. The study compares AZD0780 to a placebo and monitors participants from randomization until the primary analysis censoring date, followed by a final study closure visit. Participants will be randomly assigned to receive either oral AZD0780 or an oral placebo once daily. The treatment period lasts until the primary analysis censoring date, after which a study closure visit will occur. The study is event-driven and designed to assess the time to the first major cardiovascular event during treatment. During the study, participants will be closely monitored with various assessments to evaluate cardiovascular outcomes and safety over approximately 54 months. Researchers will track the time to first event of any component of MACE-PLUS and collect data to assess the effect of AZD0780 compared to placebo. The study includes regular visits and evaluations to ensure participant safety and adherence to treatment.

Researchers are evaluating the safety and effectiveness of HLX22 combined with trastuzumab and chemotherapy as the first treatment for patients with HER2-positive locally advanced or metastatic adenocarcinoma of the gastric or gastroesophageal junction. This phase 2, double-blind, randomized, and multiregional study compares this combination against trastuzumab and chemotherapy with or without pembrolizumab. The study aims to measure how well the treatments work in controlling the disease and improving survival for up to five years. Participants will be randomly assigned to one of two groups. One group receives HLX22 at 15 mg/kg every three weeks along with trastuzumab, chemotherapy (XELOX regimen), and possibly a placebo for pembrolizumab. The other group receives a placebo for HLX22 plus trastuzumab, chemotherapy (XELOX), and possibly pembrolizumab every three weeks. Treatment continues until the disease worsens, unacceptable side effects occur, withdrawal of consent, or other protocol-specified reasons. Throughout the study, participants will undergo regular assessments including tumor scans reviewed by an independent committee to evaluate progression-free survival and overall survival over up to five years. Other evaluations include safety monitoring and organ function tests. The study tracks how long patients live without disease progression and overall survival, aiming to better understand the benefits and risks of HLX22 combined with current standard treatments.

Researchers are evaluating ziltivekimab as a treatment for people living with heart failure and inflammation. This Phase 3 study compares ziltivekimab to a placebo in participants with heart failure who have mild to preserved ejection fraction and systemic inflammation. The study aims to assess the effect of ziltivekimab on cardiovascular death, heart failure hospitalization, or urgent heart failure visits over a period of up to 4 years. Participants will receive monthly injections of either ziltivekimab or a placebo using a pre-filled syringe or a pen-injector. The study medication is administered subcutaneously once a month for up to 4 years. The trial includes up to 20 clinic visits during which participants will be monitored and assessed. During the study, participants will use a study app on their phone to record all injections and complete questionnaires. Researchers will monitor participants for key outcomes like cardiovascular events and heart failure episodes from the time of randomization until the end of the study. Researchers are evaluating the effectiveness and safety of combining inavolisib with a cyclin-dependent kinase 4 and 6 inhibitor (CDK4/6i) and letrozole compared to placebo plus CDK4/6i and letrozole. This study focuses on participants with endocrine-sensitive PIK3CA-mutated hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced breast cancer. It aims to assess treatment outcomes in the first-line setting for this specific breast cancer type. Participants will be assigned to receive either oral inavolisib once daily or a matching oral placebo once daily. All participants will also receive a CDK4/6 inhibitor on either Days 1-21 or Days 1-28 of each 28-day cycle, along with daily oral letrozole. This randomized, double-blind study will compare these two treatment combinations to monitor differences in disease progression and safety. Throughout the study, researchers will evaluate progression-free survival from the time of randomization until disease progression or death, up to 7 years. Researchers are evaluating the effectiveness and safety of a combination treatment called triple therapy, which includes bempedoic acid, ezetimibe, and either atorvastatin or rosuvastatin. This study focuses on patients with primary hypercholesterolemia or mixed dyslipidemia who are at high or very high cardiovascular risk. The goal is to understand how well this combination lowers LDL cholesterol (LDL-C) in a real-world clinical setting. The study observes patients who have already started triple therapy within the last four weeks. No drugs are administered as part of this study; instead, it monitors the ongoing treatment with bempedoic acid combined with ezetimibe and either rosuvastatin or atorvastatin. The study measures LDL-C changes from baseline to eight weeks after starting triple therapy and continues follow-up for one year to assess lipid goal achievement, adherence to therapy, treatment changes, laboratory value shifts, and occurrence of cardiovascular events. Participants will have their LDL-C levels and other lab values assessed at baseline, eight weeks, and one year after starting triple therapy. Researchers will collect data on adverse events, adherence to treatment, and cardiovascular outcomes such as heart attack, stroke, death from cardiovascular causes, and coronary procedures during the follow-up year. The study also tracks treatment pathways and changes over this period to better understand real-world use and effectiveness of this triple therapy approach.

Researchers are evaluating the study medicine PF-08046054 compared to the standard chemotherapy drug docetaxel in adults with non-small cell lung cancer (NSCLC) that has spread or cannot be removed with surgery or radiation. Participants must have PD-L1 expression on 1% or more of their tumor cells and have experienced cancer progression during or after treatment with PD-L1 or PD-1 inhibitors, platinum-based chemotherapy, and targeted therapies for those with known genetic mutations. The trial is a Phase 3 randomized study to better understand how well PF-08046054 works alone compared to docetaxel alone. Participants will be randomly assigned to receive either PF-08046054 or docetaxel. Those in the PF-08046054 group will get intravenous (IV) infusions twice every 21-day cycle, while those in the docetaxel group will receive one IV infusion every 21 days. The treatment period may last up to 5 years if their NSCLC responds to the therapy. No other treatments are combined during the study period. Throughout the study, participants will have regular clinic visits for evaluations and monitoring to see how they respond to the treatment. Researchers will collect information on overall survival over approximately 5 years. They will also monitor safety and disease progression during these visits to understand the long-term effects and benefits of the treatments.

Researchers are conducting a pilot, observational, retrospective study across multiple centers in Italy to collect and analyze data on patients with relapsed or refractory Multicentric Castleman Disease (MCD) who have been treated with siltuximab. The study aims to provide valuable information on how siltuximab is used, its effectiveness, and safety profile in real-life clinical practice since its approval. This is important because more data are needed to improve understanding of this rare disease. The study involves gathering patient data from medical records of those who received at least one dose of siltuximab as part of their standard care from July 2016 to April 2022 at 31 selected Italian centers. As an observational and non-interventional study, no additional treatments or procedures will be applied beyond what is typically done in clinical practice. Participants' clinical information already collected during routine care will be reviewed without extra interventions. Researchers will measure the overall response rate after siltuximab infusion to evaluate outcomes. Since the study is retrospective, there is no direct follow-up or treatment administration during the study period. The goal is to better understand the real-world use and safety of siltuximab in treating relapsed or refractory MCD.

It focuses on patients with relapsed or refractory Diffuse Large B Cell Lymphoma (DLBCL) who have been treated with a combination of tafasitamab and lenalidomide. The purpose is to assess the effectiveness and safety of this treatment combination as used in real-world clinical practice under the Named Patient Program (NPP). The study collects patient data from medical records of those who received at least one dose of tafasitamab with lenalidomide between April 2022 and December 2022. Since this is a non-interventional study, no additional treatments or procedures are applied beyond what is normally done in clinical care. The treatment details and outcomes reflect standard care settings without extra interventions for the study. Participants will not undergo any procedures outside their usual care. Researchers will gather clinical variables that are typically recorded during routine practice to evaluate outcomes. The main outcome measured is the overall response rate (ORR) to the treatment combination, tracked through study completion, averaging about two years. The study monitors effectiveness and safety by reviewing existing health records without additional patient visits or tests.
