Researchers are evaluating the effectiveness and safety of oral AP1189 combined with methotrexate in adults with early rheumatoid arthritis who have not previously been treated with disease-modifying anti-rheumatic drugs DMARDs. This phase II, randomized, double-blind, placebo-controlled study aims to understand how different doses of AP1189 40, 70, or 100 mg impact disease activity and inflammation over 12 weeks in participants with active early rheumatoid arthritis. Participants will be randomly assigned to one of four groups, each receiving daily oral treatment for 12 weeks AP1189 at 40 mg, 70 mg, or 100 mg doses alongside methotrexate, or a placebo alongside methotrexate. The study compares the effects of these treatments on rheumatoid arthritis symptoms and inflammation to determine the best dose of AP1189 when combined with methotrexate. During the study, participants will attend scheduled visits for assessments including joint evaluations, blood tests for inflammation markers such as C-reactive protein CRP, and other safety monitoring. Researchers will measure changes in the Disease Activity Score 28 DAS28-CRP at 12 weeks to evaluate treatment impact. Participants will be monitored closely for safety and tolerability throughout the trial period, which lasts about 12 weeks.

Researchers are evaluating oral icotrokinra as a treatment for adults and adolescents with moderately to severely active ulcerative colitis, a chronic inflammatory disease of the large intestine causing ulcers in the colon lining. The study aims to assess how well icotrokinra works, along with its safety and tolerability in this population. This is a Phase 3, randomized, double-blind, placebo-controlled trial with a parallel group design including both adults and adolescents. Adult participants will be randomly assigned to receive either icotrokinra or placebo daily by mouth during a 12-week induction phase. At Week 12, those showing clinical response will enter a maintenance phase where they will continue icotrokinra or placebo daily for 40 weeks. Adults who do not respond will also enter the maintenance phase and receive icotrokinra. Adolescents will receive open-label icotrokinra during induction and then continue on icotrokinra during maintenance regardless of response. After completing the 40-week maintenance phase, eligible participants may join a long-term extension study. Participants will be monitored regularly through clinical assessments at specified time points including Week 12 for induction and Week 40 for maintenance. Outcomes measured include rates of clinical remission, symptom improvement, endoscopic and histologic healing, and quality of life scores. Safety will be evaluated by tracking adverse and serious adverse events throughout the study. The total study duration may extend up to approximately 6 years, ending in 2032, allowing long-term evaluation of icotrokinra in ulcerative colitis management.

Researchers are evaluating efruxifermin EFX in a phase 3, randomized, double-blind, placebo-controlled study involving adults with compensated cirrhosis caused by NASH Nonalcoholic Steatohepatitis or MASH Metabolic Dysfunction-Associated Steatohepatitis. This study aims to assess the safety and effectiveness of EFX in preventing significant clinical events such as disease progression and liver decompensation over a period of up to 5 years. Participants are randomly assigned to receive either efruxifermin 50 mg or a placebo, both given by subcutaneous injection. The study includes two cohorts one with biopsy-proven compensated cirrhosis and specific metabolic scores, and another with biopsy-proven or non-invasive diagnosis of compensated cirrhosis. The study treatment and monitoring extend up to 5 years, with evaluations at 96 weeks and long-term follow-up to track liver fibrosis, markers of liver injury, insulin sensitivity, glycemic control, body weight, and safety outcomes. Researchers are evaluating the effects of a triple therapy inhaler combining budesonide, glycopyrronium, and formoterol fumarate BGF MDI 32014.49.6 g compared to a dual therapy inhaler with glycopyrronium and formoterol fumarate GFF MDI 14.49.6 g on heart and lung outcomes in adults with Chronic Obstructive Pulmonary Disease COPD who have a higher risk for heart and lung events. This Phase III study is randomized, double-blind, and conducted at multiple centers, focusing on participants with COPD and elevated cardiopulmonary risk. Participants will receive either the triple therapy inhaler or the dual therapy inhaler, both administered twice daily. The study compares these two inhalers over a period of up to three years, monitoring for serious cardiac or COPD events. This research aims to evaluate the antiviral effects of S-337395 compared with a placebo in adults who are not hospitalized but have respiratory syncytial virus RSV infection and are at high risk of progressing to severe disease. Participants must start treatment within 72 hours of showing RSV symptoms. The study is a Phase 2b trial and involves adults with specific risk factors such as older age and chronic lung or cardiovascular disease. Participants will be randomly assigned to receive either a high dose or low dose of S-337395, or a matching placebo. The treatment is given orally twice daily for up to 5 days. The study is double-blind, meaning neither participants nor researchers know which treatment is being administered during the trial. This Phase 3 study aims to compare KarXT with a placebo to see how well it works in treating psychosis symptoms associated with AD, focusing on changes in hallucinations and delusions. Participants will receive either KarXT capsules at varying doses or placebo capsules in a randomized, double-blind setup. Researchers are studying the drug JNJ-88545223 to see how well it works compared with a placebo in adults who have active psoriatic arthritis PsA. This study aims to determine if JNJ-88545223 can reduce the signs and symptoms of PsA and improve the health of joints and skin. The trial is a randomized, double-blind phase 2b study designed to evaluate different doses of the drug. Participants will be randomly assigned to receive one of four treatments from Week 0 to Week 16 placebo, or one of three doses of JNJ-88545223. Each group will receive their assigned treatment under blinded conditions to compare the effects. The study focuses on evaluating the efficacy and safety of these doses over the 16-week treatment period.
