Study of Blood Fat and Inflammation Markers After Oral Fat Test to Predict Diabetes Risk and Complications in Adults Aged 50-70

Researchers are investigating the response of triglyceride-rich lipoproteins and inflammatory cytokines after an Oral Fat Loading Test OFLT in people with type 2 diabetes T2D, prediabetes, and healthy controls. The study aims to understand if these responses can serve as early markers for the risk of diabetes progression and related complications. This observational cohort study includes three groups individuals with T2D, prediabetics, and control subjects. Participants will undergo the OFLT, which involves consuming a fat-rich, sugar-free mascarpone cream after a 12-hour fast. Blood samples are collected before and at 1, 2, 3, 4, and 6 hours after the fat intake to measure triglycerides, lipoproteins, cytokines, and other markers. The study includes a cross-sectional phase followed by a 36-month prospective follow-up to assess whether these biomarkers predict progression to diabetes, nephropathy, and major cardiovascular events. During the study, participants will have multiple blood draws to assess changes in triglyceride levels, lipoprotein particles, and inflammatory markers. Researchers will monitor progression from prediabetes to diabetes and the development of kidney and heart complications over three years. The main outcomes include triglyceride concentrations after OFLT and diabetes progression, with secondary measures including various lipoprotein and cytokine levels. Participants will be followed regularly to evaluate these markers and health outcomes.
