What this Trial Is About
Alzheimer's disease (AD) is a leading cause of dementia and is expected to affect over 100 million people by 2050, presenting a major public health challenge worldwide. AD involves progressive memory and cognitive decline linked to specific brain changes, including amyloid plaques and tau protein tangles. The disease progresses through stages from preclinical with normal cognition but positive biomarkers, to mild cognitive impairment (MCI), and finally to dementia, which disrupts daily activities. This research focuses on subjective cognitive decline (SCD) as an early symptom and aims to better understand the clinical and biological changes leading to dementia. This observational study involves cognitively healthy individuals with SCD, people with MCI, and those with mild dementia. Participants are assessed using questionnaires measuring their subjective perception of cognitive decline, including self and relative reports. The study explores how these perceptions change over time and relate to cognitive performance, emotional patterns, and Alzheimer's biomarkers. There is no intervention or treatment given as part of the study. Participants will engage in long-term follow-up assessments including the Subjective Cognitive Decline Questionnaire both at the start and during the study. Researchers will evaluate changes in subjective complaints alongside cognitive and affective measures, as well as biomarker data when available. The goal is to identify early indicators of Alzheimer's progression and understand how perceptions of decline evolve, involving both participants and their relatives. The study is expected to continue through 2027.
