What this Trial Is About

Researchers are evaluating the antibacterial effectiveness of two different intracanal medicaments, Bio-C Temp and UltraCal XS, in mandibular molar teeth diagnosed with chronic apical periodontitis. This randomized controlled clinical trial focuses on reducing bacterial load inside root canals during primary root canal treatment. The study includes systemically healthy adults aged 18 to 65 years who have radiographic evidence of periapical radiolucency and require root canal therapy. Participants are randomly assigned to receive either Bio-C Temp, a calcium silicate-based bioceramic medicament, or UltraCal XS, a calcium hydroxide-based medicament. Both groups undergo standardized root canal shaping with heat-treated nickel-titanium rotary files and irrigation using sodium hypochlorite, EDTA, and passive ultrasonic activation. The assigned medicament is placed inside the root canals 1-2 mm short of the working length and sealed temporarily for 14 days before removal and further irrigation. During the study, bacterial samples are collected at several stages before treatment, after root canal preparation, after medicament removal, and after final irrigation. The bacterial load is measured using droplet digital PCR to compare the two medicaments antibacterial effects. Pain levels are recorded with a visual analog scale. After sampling and irrigation, root canals are filled and restored, with final radiographs taken to assess treatment. Participants are monitored throughout the treatment process, which lasts about two weeks from medicament placement to the second appointment.
